چکیده

A new enzyme activity that resembles terminal deoxynucleotidyl transferase (TdT-like activity) was found in rat liver. Upon treatment of rats with a hepatocarcinogen, N-2-fluorenylacetamide, the TdT-like activity increased in parallel with DNA polymerase Ci and 'Y. By phosphocellulose column chromatography, the TdT-like activity was separated from DNA polymerase Ci, fJ, and 'Y. With a poly(dA)-initiator, dGTP or dCTP was the most efficient substrate in the presence of Mg2+ or Mn2+, respectively. In the presence of Mg2+, the enzyme utilized poly (dC) as an initiator most efficiently. The TdT-like activity differed from previously described terminal deoxynucleotidyl transferase (E.e. 2.7.7.31.) in its Km value for dGTP and in its sensitivity to ATP or to an antibody against calf thymus terminal deoxynucleotidyl transferase.

Protective Effect of Folic Acid against Apoptosis Induced by Ischemia/Reperfusion Injury in Rat Liver

The anti-apoptotic gene bcl-2 is located in mitochondria, but it is uncertain whether its expression affects hepatocyte survival in ischemia/reperfusion (I/R) injury. This experiment was designed to evaluate the role of folic acid in expression of bcl-2 in I/R in rat liver. Eighteen Wister rats were divided into sham-operated controlgroup (C) (n=6), I/R group (...

Track detection on the cells exposed to high LET heavy-ions by CR-39 plastic and terminal deoxynucleotidyl transferase (TdT)

Background: The fatal effect of ionizing radiation on cells depends on Linear Energy Transfer (LET) level. The distribution of ionizing radiation is sparse and homogeneous for low LET radiations such as X or γ, but it is dense and concentrated for high LET radiation such as heavy-ions radiation. Material and Methods: Chinese hamster ovary cells (CHO-K1) were exposed to 4 Gy Fe-ion 2000 keV/...

Increased activity of polynucleotide ligase from rat hepatoma induced by N-2-fluorenylacetamide.

Polynucleotide ligase in both the soluble fraction and the nuclear extract, which catalyzes the covalent joining of 2 segments of an interrupted strand in a DNA duplex, was studied with normal rat liver and hepatoma induced by yV-2-fluorenylacetamide. The activity of this enzyme in hepatoma was about 5 times greater in the soluble fraction and about 3 times greater in the nuclear fraction than ...

Normal rat liver hepatocytes and the hepatocytes of hepatic nodules induced by N-2-fluorenylacetamide respond to 70% hepatectomy with vigorous mitotic activity. The administration of L-asparaginase at the time of operation

Normal rat liver hepatocytes and the hepatocytes of hepatic nodules induced by N-2-fluorenylacetamide respond to 70% hepatectomy with vigorous mitotic activity. The administration of L-asparaginase at the time of operation delays the mitotic cycle of every normal hepatocyte by 10 to 12 hr. When L-asparaginase was administered to rats which had received N-2-fluorenylacetamide, an 80% inhibition ...

Zataria multiflora Essential oil Prevent Iron Oxide Nanoparticles-induced Liver Toxicity in Rat Model

Over loading of iron oxide nanoparticles can causes the liver injury through overproduction of free radicals. Zataria multiflora Boiss. (Lamiaceae) has been used for many years in folk medicine due to its antioxidant and antibacterial activities. This study evaluates -for the first time- the effect of Z. multiflora essential oil (EO) against iron oxide nanoparticles hepatotoxicity in rat model....
